What is GHRP-6 acetate?

This listing is GHRP-6 acetate: 5 mg of growth hormone releasing hexapeptide in acetate salt form, made by PEPTYVA in Mexico. GHRP-6 is a synthetic peptide that stimulates growth hormone (GH) release. The vial is sold for laboratory research only; it is not a medicine and must not be used in humans or animals.

Its sequence is His-D-Trp-Ala-Trp-D-Phe-Lys-NH2, with D-tryptophan at position 2 and D-phenylalanine at position 5. It is among the older GH-releasing peptides, with endocrine studies in healthy men published in 1990.

How is GHRP-6 studied in research?

GHRP-6 is a classic tool for the growth hormone secretagogue receptor (GHS-R). The 1990 human study already proposed that its GH-releasing activity reflected a separate physiological system. In 1996, researchers cloned this G protein-coupled receptor from the pituitary and hypothalamus of swine and humans and showed that it was the target of the synthetic secretagogues. Its natural ligand, ghrelin, was identified in 1999.

Because it acts through this separate pathway, GHRP-6 is used to study how GH release is controlled independently of GHRH. Animal research has also examined links to feeding behavior: in a 1995 rat study, centrally given GHRP-6 was followed by eating, and the eating response did not track changes in plasma GH. These are rodent observations and do not describe effects in people.

What does the current evidence show?

Human data for GHRP-6 are short endocrine measurements in small groups. In a 1990 study of 18 healthy men, GHRP-6 raised serum GH. Prolactin and cortisol rose about twofold only at the highest level tested, and LH and TSH did not change in the first hour.

A 1992 study in five healthy men showed that the GH response to GHRP-6 persisted after the pituitary had been made unresponsive to GHRH, while the response to GHRH itself was lost. The authors concluded that the acute effect of GHRP-6 does not depend on endogenous GHRH.

These studies measured hormones over hours. Efficacy for any use in people has not been established, and FDA notes possible effects on cortisol and blood glucose.

How does GHRP-6 differ from GHRP-2?

Both are hexapeptides that act on the same receptor, but their sequences and status differ. GHRP-6 begins with histidine and D-tryptophan, while GHRP-2 begins with D-alanine and D-2-naphthylalanine. GHRP-2, as pralmorelin, holds a Japanese approval as a diagnostic agent. For GHRP-6 we found no approved product in Drugs@FDA. GHRP-6 is also slightly heavier, at 873.0 g/mol as the free base.

What is the regulatory status of GHRP-6?

GHRP-6 is not an approved medicine in the US, and Drugs@FDA lists no product containing it. On September 29, 2023, FDA placed GHRP-6 in 503B Category 2, noting limited safety information, a risk of immunogenicity and concerns about cortisol and increased blood glucose due to lower insulin sensitivity. In Mexico, this vial is sold as research material only.

Outside medicine regulation, anti-doping laboratories have published methods to detect GHRP-6 and its main metabolites in human urine, which reflects how closely its circulation is monitored.
